چکیده

In the presence of 0.15% oxgall, the B-galactosidase activity of whole cells of Lactobacillus bulgaricus was increased. However, higher concentrations of oxgall decreased the enzyme activity. In the absence of oxgall, little or no B-galactosidase was exhibited by the whole cells. When the cells were lysed to obtain cell-free extracts prior to assay, there was some decrease in activity of the enzyme in the presence of 0.15% oxgall. Increasing concentrations of oxgall did not further reduce the activity. Apparently, the oxgall increases the permeability of the cells upto a certain concentration (0.15%) to allow more substrate to enter, resulting in more hydrolysis of the substrates. However, at concentrations higher than that, the enzyme activity was inhibited by the oxgall.

Non-fusion and fusion expression of beta-galactosidase from Lactobacillus bulgaricus in Lactococcus lactis.

OBJECTIVE To construct four recombinant Lactococcus lactis strains exhibiting high beta-galactosidase activity in fusion or non-fusion ways, and to study the influence factors for their protein expression and secretion. METHODS The gene fragments encoding beta-galactosidase from two strains of Lactobacillus bulgaricus, wch9901 isolated from yogurt and 1.1480 purchased from the Chinese Academy...

Homodimeric β-Galactosidase from Lactobacillus delbrueckii subsp. bulgaricus DSM 20081: Expression in Lactobacillus plantarum and Biochemical Characterization

The lacZ gene from Lactobacillus delbrueckii subsp. bulgaricus DSM 20081, encoding a β-galactosidase of the glycoside hydrolase family GH2, was cloned into different inducible lactobacillal expression vectors for overexpression in the host strain Lactobacillus plantarum WCFS1. High expression levels were obtained in laboratory cultivations with yields of approximately 53000 U of β-galactosidase...

Hydrolysis of lactose by immobilized microorganisms.

Cells of Lactobacillus bulgaricus, Escherichia coli, and Kluyveromyces (Saccharomyces) lactis immobilized in polyacrylamide gel beads retained 27 to 61% of the beta-galactosidase activity of intact cells. Optimum temperature and pH and thermostability of these microbial beta-galactosidases were negligibly affected by the immobilization. Km values of beta-galactosidase in immobilized cells of L....

Characterization of Lactobacillus bulgaricus Bacteriophage ch2.

Bacteriophage ch2, a virulent bacteriophage of Lactobacillus bulgaricus CH2, was characterized according to its morphology, genome size, structural proteins, and growth kinetics. Electron micrographs revealed that bacteriophage ch2 has an icosahedral head of 50-nm diameter and a long tail of 170 nm. Its genome is linear and 35 kilobases in length, and its structural proteins consist of two majo...
